Suggestions for more sophisticated synth-sounds for use with my midi-sequencer?

I don't know what the possibilities of those are, but I know in Ableton Live 8, there is an instrument that you put a sample in it, and it will play like an instrument. You can get great sounding stuff with little adjustment.
 
I use Master Tracks Pro 6.8.4 as my sequencer. PC-based, Windows XP Home Service Pack 2, I am looking for more sophisticated sounds. In my midi- set up I have the choices of : SB Audigy 2 Synth A, SB Audigy 2 SW Synth, SB Audigy 2 Synth B , and Microsoft GS Wavetable SW Synth.
I am only interested in soft -synths. I don't want to have to mess with a bunch of midi-cables, and my space is limited.
