plate culture of bacteria? Which of the following steps are NOT done for preparing a slide from an agar plate culture of bacteria? [Check ALL that are NOT done]

a. Add a 1/2 drop of distilled water to slide.
b. Flame loop before sampling from plate.
c. Use a coverslip to hold bacteria in place.
d. Transfer several "loopfulls" of bacteria to slide
e. Spread sample over slide and fix.

'D' would probably be the best choice because several "loopfulls" may be too thick to properly visualize the bacteria under a microscope. Generally, one loopful evenly spread throughout the drop of water will work fine.

You can do all of these things to prepare a slide of bacteria that are being sampled from a culture on an agar plate.
