What test should I have done for my pregnancy? mothers and expectant mother only?

On Monday I am having a doctors appointment. We are going to start test, but I want mother who have already had kids, which test do you think are the most important?
Yes I'm talking about down syndrom,
 
with each tube of blood they are able to do about three tests, so they will test you for whats needed...
