Silent Movie - one word spoken in the whole film (by Marcel Marceou) and the fly in the soup gag got a whole cimema to burst out laughing.

Others are the Holy Grail, Blazing Saddles, Airplane and the Pink Panther (Peter Sellers version - Does your dog bite...?)
 
I think that Monty Python And The Holy Grail is one of the best films ever, not merely one of the funniest. Second to that the only film that has had me almost crying laughing is What's Up Doc, and its frenetic madness right from the start which never lets up. The sight of the runaway Chinese Dragon careering down the San Francisco streets always has me creased up.
