PHP updating the 40 in this line?

In PHP there is autotrigger in 5.1 version
I have the field which is skills_level " 0,0,0,0,0,0,0,0,0,1,0,3,0,40,0,0,0" the data is this
i want to update the 40, how would i do this?
This is for MySQL by the way
