need help regarding plant tissue culture?

I want to tissue culture Gerbera plants so I made a media containing agar sugar vitamin auxins and cytokinins.Adjusted the PH to 6. Then I sterilized my explants using Alcohol Hydrogen Peroxide and Bleach. Placed them under fluorescent lights at a temp of 28 C. After 15 days bubbles started to form around the explant in the media which kept growing with the passage of time.In a couple of weeks the mass of bubbles turned green completely. At this stage I had a power failure for almost 36 hours. So the bubbles started to disappear and eventually all the bubbles were gone. I want to know whether it was due to the power failure that my experiment collapsed? The formation of green bubbles is a sign of success or not? Actually I m a novice to tissue culture so I know nothing about it.
 
Yes power failure for that long can cause some problems with tissue culture. The callus reacted to the darkness and that can cause the callus formation to shrink or die. That is why we have a back up system just in case if the power went out. Sorry to hear that. You will might have to start over again.
