My friends have booked their wedding in Mexico...?

Just simply restate the reasons you gave in the first place, since nothing has changed. You still do not know your Christmas plans, and you still do not think you will have enough vacation time saved up. Don't feel bad; it's their fault for not checking with friends if they expected them to attend. If they are true friends, they will understand.
 
...for a week in December and I live and work in Europe. I was invited and I said that I could not commit since I didn't know my plans for Christmas, nor how much vacation I would have left, nor what would be happening with Swine Flu in December.

Now they've asked both my spouse and I to be part of the wedding party. In their words, they did this to encourage us to attend despite our misgivings.

This seems like a huge imposition to me. I believe they are being so insistent because they booked their wedding before bothering to check if anyone would be able or willing to come.

How do I say no??
 
Simple, Hon..you repeat your Mantra '' I'm sorry, but I don;t know yet my family plans for Christmas, how many vacation days I would have left, and the flu situation so it's best you ask another couple. Perhaps the prudent thing on your part would have been to check with your potential guests availability BEFORE you booked your day. I'm sorry, I just can't commit right now "

You can say no. It's simple "I'm sorry, we just can't commit at this time"
