Is it ok to argue against the question in an essay?

I have a synoptic paper to do and I have been asked which theme I thought my university course was mostly about. Could I argue against this and say that I believed it focused on three central themes? It's a 2000 word paper so I'm worried it would be hard to write that length on one topic which is quite limited.
