Is an upgrade from a ninja 250 to a ducati 848 too much of a jump?

not at all. Neither bike is that heavy or or hard to control. As with any bike ride within your comfort level and you will be fine. Just because you have a bike that goes 150MPH doesn't obligate you to drive 150MPH. If that is what you want then why waste your money buying inbetween bikes just get the one you want.
 
Sort of a big jump, you may want to consider a DucK Monster 669 if you want Ducati but there are other bikes in the 600 class that may fit your butt better,

If you really think you're ready for the 848, ask your insurance agent how much your rate will be jacked up. It may make you rethink your option.
