In US, Can I buy medicine at the grocery or in a supermarket 24/7?

Those over-the-counter medicine aisles are really opened 24/7 without pharmacist management?
Then what if you want to talk to pharmacist to find the most appropriate medicine?
And I heard that NOT all the drugstores (like CVS) have pharmacists. Is it true?

Please let me know what it's like in every state possible. That'll be very helpful for me.
