In RENT, why in the beginning of the Tango: Maureen did Mark say 'Anything but that'?

In the beginning of the song the Tango: Maureen in the RENT movie, the beginning was like this:
Mark: Say something, anything,
Joanne: Test, one, two, three.
Mark: Anything but that.
Why does Mark have a problem with Joanne saying "Test one, two three"? I'm assuming it has to do with the fact a majority of Mark's friends have STDs ('test') but I'm not completely sure...I haven't seen the full play/movie only heard most of the songs and I know the plot. You don't have to worry about spoilers; I know this is an odd question but someone please answer it, I'm curious! Thank you.
