How to properly do a throat swab?

In my medical microbiology class, we have to do throat swabs. What's the proper way of doing a throat swab?

I've been told three different methods:
1) Touch the back of the throat
2) Touch the right tonsillar pillar and follow up to the uvula on to the left side
3) Touch the area in front of the uvula

Which method is correct?
