How to lose weight with exercise bike? RPM, resistance?

My sister just gave me an exercise bike, I was wondering what is the best way to use it to lose weight? I'm not really overweight- 5'8", 140 lbs- but I do need to lose about 10 pounds and improve my health a bit.
I am planning on using it for like 30-45 minutes a day or a bit more. I'm just confused about the RPM and resistance stuff. What are the best settings/levels for those if you want to lose weight? I have seen some things saying to go fast like low resistance/high RPM but others say to do high resistance/low RPM at a steady slower pace. And will higher resistance really make your leg muscles a lot bigger if you're female?
