Why? The whole idea of plywood and "concrete" backer boards is to give you a surface that "won't move(thereby cracking the tiles).
Seeing the floor is concrete already, you already got the "backer board" in place. Gravity will hold the tiles down. Ceramic tile glue sticks like cement"to anything". So the paint can stay. Just use a 4" spackle knife to lift up any obviously flaking paint.. However, did you check to see that your floor is 100% flat? Using a long straight edge.48-96" long. You may have to "float the floor first" with a thin slurry of concrete to give you a perfectly flat surface. That way the tiles won't break because they are supported completely.
Then go for it. Tiling is the same around swimming pools which are concrete patio's. Paint is not a concern. Make sure to grout the grooves afterwards.
