Depends on where you go. Conventional places can take up to two weeks. Places like LensCrafters, that have the lab in the same building as the optometrist's office, can do it in about an hour, unless they have to special order the lenses for your frames (which is why I love LensCrafters... In the 20 years I've worn glasses, I've only had to wait once for a pair of glasses).
