How do you use a directory as a php variable?

I am in the middle of creating a small little project and wanted to have user profiles work like this:
A good example would be Twitter.
How does "http://twitter.com/KaiserCoaster" get the variable for "KaiserCoaster"?
I know its not a page, but I don't know how to do that. Any help?
