I mainly use many layers in Photoshop to get all my desired effects. One layer for your pencil lines, one layer for your colors (hair layer, clothing layer, ect..) then your shading layer.
You can find easy effects to use in the plug-ins and also I like to use styles to create my effects.
You need to play around with all the options.

I say the best way for you to get it the way you want is to treat your drawing as if they are present in the real world.
Tell yourself where is the sun, where would the shadows cast,
what angle is the camera, what kind of lighting do i have, Build up your mood and set a story line along with it.
this helps me when I draw and create.
Also pick up books on your genre. Like Anime or still life or whatever type of effect you are aiming for.

Good luck hope this helps
I am currently working on my website, but i create it all in photoshop
