How do I fix a flat spot on my bicycle tyre?

Whenever I am riding along on a smooth road I notice that each time my wheel revolves I feel a bump. It is caused by a flat spot on the tyre of the bicycle wheel. Does anyone know what is causing the flat spot and how to fix it? I Have already deflated and reinflated the inner tube several times and had no luck, I have even taken the tyre off again to check that the tape that goes over the spoke screws is even throughout the wheel. But there is still a flat spot.
 
This flat spot is on the wheel not on the tyre! The wheel needs to be inspected for true and then trued accordingly. Any good bike shop will have to do this for you, doesn't sound like you have the know how to preform this task!
