My wife has been asking me to be rougher with her in bed and I feel really uncomfortable.

I've done some outrageous things with women before, but they weren't my wife.

To top it off, I'm a big guy and she's really small and I'm afraid of hurting her.

I'm also afraid of crossing the line, because she has been sexually assaulted before and that has come up during sex before.

umm you could try tying her to the bed... maybe blindfolding her or putting something over her mouth so she cant talk... just take control and feel empowered by it!
 
Read a bit about soft-BDSM-ish stuff.
Some tying up, stop spanking while doing it, holding her wrists together, or calling her names. Gagging, blindfolding. Softly holding her throat while doing it, without completely choking her. Just give her a safe word, to use if it's going out of hands, and explore things.
