If the wheel is locked then chances are that when you repositioned it it is not in correctly. Did you put all the spacers back in their appropriate places? Did you get a new tire or a new wheel? I strongly suspect the wheel is slightly cocked and that is why it is not moving also why the brake is not working correctly. Also anytime yoy remove/reinstall a wheel from a bike, the brake has to be primed-ie pump the lever until the pressure rebuilds and holds the pads next to the brake rotor.
