ford f-250 cylinder #2 misfire code?

my f-250 engine has started to run a bit rough in the higher gears especially under load and uphill. It seems to go through 1/2/3 fine but when i get to a speed and coast and then re-accellerate it misses a bit. I had autozone scan and they had a pending code for cylinder #2 misfire. my check engine light is NOT on. I just bought the truck from a used care dealer with 175,000. It drove home fine and on my second tank of gas this started. I thought it may be water in the fuel system as it was beyond below E when i got it so i am running a fresh tank with some STP cleaner through it. Do you think I may have simply a bad plug or wire? Any ideas as for what I can check. I am pretty handy although no mechanic and I am just starting a business so I have no $$$. Thanks!
how do i go about checking the plug/coil?
 
if its the 5.4l it has coil on plug, which means there is a coil on each plug and has a spark plug boot that goes between the coil and the plug, so there is really no ignition wires. it they are the original plugs i would get them changed if not the coil it self is probably bad.
