I presume you don't really mean "like Harry Potter" because there _is_ no other fandom that big. Twilight's a tenth the size. And, while it's not a book, Naruto is bigger than Twilight (by fanfics posted on fanfiction.net) and is a fantasy series by a Japanese author with Japanese characters.

I don't see why not. I don't read books based on what colour the author's skin is. Maybe it'll be Noughts and Crosses by Malorie Blackman. It's certainly good enough to deserve it.
 
If it's a really good story idea then I don't see why not. Not all the characters in the book have to be white and the author doesn't have to either. FYI, I don't consider Twilight to be in the same league as Harry Potter.
