Can I use a semi-gloss latex paint over a flat latex paint?

I have 2 new gallons of quality latex paint in the same color. One is flat and the other is semi-gloss. Can I use the flat paint for the first coat and the semi-gloss for the final coat without any problems? I don't know if this matters, but it will mainly be over fresh drywall. Thanks!
Oops. I forgot to add that we will be using a primer, probably a primer & sealer for new construction/fresh drywall.
