You'll have to create (subdivide) a new lot to be able to sell it. To do this, the lot will have to be in accordance with your municipal zoning laws - it will have to have sufficient size, access to a roadway (or an easement or right of way through the lot that you're going to keep), water and sewer service (or well and septic) etc. etc. If it meets your zoning requirements, then, sure, you could sell it.

As an alternative, you could sell it to another neighbor who adjoins your lot if he/she would want to annex it onto their lot.
 
This answer applies if you're in the UK. Yes, you can sell your garden to anyone who's prepared to buy it but just because it's big enough for a building plot doesn't mean that planning permission will be granted. If you sell it with planning permission you'll get more for it but of course that means you'll have to make the application for the permission which will involve some cost.

One thing you need to bear in mind: if you have a mortgage on your property your lender will have to give consent to the sale before it can go ahead.

Incidentally, if the plot's only big enough for one three-bed semi, where are you proposing that the other semi's going to go? You might want to give that some thought before you invest too much effort and money into it!
