Would elevation make a difference on a sniping shot?

For instance. If i am shooting a .243 or a .308win or whatever, whats the differnce between shooting 500yds from a roof/cliff down to the ground or from ground to the ground. My point is would elevation effect accuracy/trajectory. Is there anything to adjust or not. Like in the beginning clip in 'SHOOTER'
