When cooking a meal with several ingredients, how to determine calories per serving?

I'm new to the whole calorie counting thing. If I'm making a meal for several people, which includes several ingredients, how can I determine how many calories I am consuming per serving?

For example, say I make a stir fry with sweet and sour tofu. The pot of tofu, sauce, and vegetables is large and full of ingredients. When I scoop it onto my plate, how do I know how many calories I am ingesting per scoop or spoonful or serving or whatever?
