What to do about knee pain with climbing or going down stairs, caused by biking?

About two weeks ago I got on my bike for the first time in the spring, rode two miles, and spent most of a day drilling holes in pieces of wood braced against various parts of my body while sitting/standing/kneeling/squatting on a concrete floor in a 50 degree room. When I tried to go home my bicycle gave out on me after a quarter mile, and my body gave out on me after walking another quarter mile. I had to call a friend (to whom I am super grateful) to get a ride.

Since then I've had severe pain in my right knee when I try to go down stairs, and to a lesser extent when I climb them. It's bad in the morning when I first wake up, seems to get better as I stretch and warm up by walking around the house, and it's really bad if I come home after a longer than usual walk (more than 3/4 mile at a time or a mile and a half round trip). I haven't gotten back on my bike since then, because I haven't had time to take it to the shop. I'm probably going to take it in tomorrow.

It doesn't bother me when I walk. It seems that I don't actively use the parts involved unless I'm climbing stairs. Occasionally my right knee hurts when I'm getting up from a chair, as well. It seems that it has to be bent significantly while bearing weight to trigger the pain. After I go down the stairs I find that my right knee is warmer than my left, and a bit sore.

Does anyone who knows about these things have any recommendations for exercises, treatments etc. that I can do to strengthen my knee so it no longer gives me pain?
