The directions are right on the box.
There is no way she can get to it, or lick it off, if you put it exactly where you are instructed to put it. Personally, I put it a little higher up on the neck than "between the shoulder blades". Just a little, to make absolutely sure he can't lick it.
It will not "dry" -- it will sort of "spread" all over the body, hair by hair. This is how it works. It will be hours before it doesn't look wet anymore at that particular spot.
It does not matter if she climbs around or not - just allow her to do her normal stuff. No, you don't need to wear gloves, just be sure to wash your hands afterwards.
It's also very important to make sure you do this each month.

I just read the last question - there is NO reason in the world that this should "get all over her fur" and it is your responsibility to make sure it doesn't.
Just read the directions on the box!
Take the tube, take the top off, turn it over and "puncture" the tube. OR - snap off the end of the tube - it depends on which brand you bought (Advantage or Frontline). All the while you should be talking sweetly and gently to the kitty. Hold the tube in one hand.
With the other hand -- Now, kneel down and gently/firmly grasp the kitty behind the scruff of the neck - you don't have to GRAB her or scare her -- and while you are petting/kneading her scruff with this free hand, turn the tube over to the spot between her shoulder blades ( a little higher than that) and move the tip around to make sure you use ALL the liquid. Talk to her the whole time you're doing it, to distract her.
It only takes a moment. Once you've rubbed the tip around a bit (about the area of a dime), then just release her neck and rise up and walk away.
Maybe give her a bowl of wet food for a treat/reward.
