I recently did a practical involving subcloning of hlyA into pBluescript and while I found that hlyA was sucessfully added to the pBluescript plasmid, when added to bacteria (which when studied later to have the gene in its DNA) did not express the product of the hlyA gene.
Can anyone suggest a reason why hlyA was not expressed despite the fact the bacteria had the gene in its DNA?
