Well, you appear to have two very different questions.
The shuttle program was not the only U.S. program to send missions into space.
The Apollo and Gemini programs were first, and did not use shuttles at all.
I'm assuming you mean Human Space Flight. The numbers for every launch in the U.S. might be difficult to figure out, although I'm sure someone on this website will be willing to track down that information as well. I found a list of every HSF mission on wikipedia, but it includes missions of other nations.
To date, there have been 124 U.S. shuttle missions that have sent astronauts into low earth orbit. (5 free flight tests).
