How do I gain the interest of a literary agent as a new author?

I just finished a bio book and have send out so many inquery letters - some agents have shown some interest, but in the end they all say sorry but keep on trying, I'm sure you will find the perfect fit for your manuscript. How can I generate a better interest to get my project published??
