does hiking suppose to hurt your back?

okay so I wasnt hiking through the mountains but I was backpacking with 35 lbs in my town. I dont know if my shoulders are misaligned or if my backpack straps weren't at the same level or what. Because after 3 hours of walking my right side of my back and shoulder, all of that region ached bigtime/and or cramped. Because after the 1st hour I started feeling it and after I took it off I felt it even more. So right now it feels like its tight and if I stretch my right arm it feels like the muscles are locked. Am I backpacking wrong or do I need a better backpack. Or maybe every hiker/backpacker feels this? I want to know your feedback
I heard thats the pounds that conventional hikers carry atleast when they hike. And they burn a huge amount of calories.
 
You could try getting a better backpack if the one you have now isn't quite working for you, but also, it could be that your body isn't used to carry so much weight around.
Also, when I go hiking through the mountains, I can usually start to feel the weight of my back pack (20-35 pounds) in about 3-4 hours. I have a regular backpack not a hiking one, and I think using a hiking pack could make the trip easier.
Though, I've never tried hiking through town (on flat surfaces). I'm usually on an incline when i hike...
