Not gender, that's pretty easy.

But, sex.

I don't believe that..
They are a certain sex, and can't change it. People who want to change it can't, because there is nothing to change.
They are the sex they think of themselves, but they just happen to be in another sexes body.

Have you never heard of sex reassignment surgery? There are a myriad of surgical procedures people go through to change their sex, not to mention hormone therapy. So, yes, I believe (and know) that people can change their sex.
 
i do, because, when your developing in your mothers womb your brain decides what sex you are before your genitals develop.
and most cases in trans-genders their brain has mixed signals on what sex they are, so even though physically they are male/female their brain doesn't know (male and female brains are different to an extent)
