i am doing direct sequencing for my project. for precipitation with ammonium acetate, when we added water to the DNA after centrifuging the sample for 1 minute, do we mix the water and the DNA with pipette? i didn't do it because i thought that i shouldn't ruin the palette at the bottom of the tube.
I had done my precipitation with ammonium acetate so i did big dye sequencing. the thing is that i forgot to mix the precipitated DNA with pipette before loading it in tray for big dye but i always pushed the pipette tip to the bottom of the well to take up the precipitated DNA. is there any chance that i still got DNA (not water only)?
